Effect of methylmalonyl coenzyme A, a metabolite which accumulates in vitamin B 12 deficiency, on fatty acid synthesis.

چکیده
We have proposed that the accumulation of methylmalonylCoA, which occurs when a deficiency of B12 coenzyme exists, may lead to its incorporation into fatty acids in place of malonyl-CoA. This leads to the production of fatty acids, which are not usually produced and which may be physiologically undesirable. As a first step in the test of this hypothesis, we have examined the effect of methylmalonyl-CoA on fatty acid synthesis in vifro, in rat liver supernatant fractions. It was shown that methylmalonyl-CoA inhibits fatty acid synthesis and is incorporated into fatty acids.

متن کاملThe effect of vitamin B12 deprivation on the enzymes of fatty acid synthesis.
The enzymes of fatty acid synthesis from liver and brain in normal and Blz-deprived rats were studied. Both total and specific activities of fatty acid synthetase and acetyl coenzyme A carboxylase were 2to 5-fold greater in Blz deprivation than in the normal state. The presence of excess activators in the BIp-deprived rat or an excess inhibitor in the normal rat was not found by serial admixtur...
متن کاملAn effect of vitamin B 12 deficiency in tissue culture.
Rat glial cells were grown in tissue culture in vitamin BW deficient medium. As the intracellular level of coenzyme Bls decreased, the cells made increasing amounts of two unusual fatty acids. The fatty acids were unbranched acids with 15 and 17 carbon atoms and arose as the cells progressively lost the ability to metabolize propionic acid through the BIz-requiring methylmalonyl-CoA mutase reac...
متن کاملVitamin B-12.
V itamin B-12 is a cofactor for 2 enzymes. In the cytoplasm, methionine synthase requires vitamin B-12 in the form of methylcobalamin and catalyzes the conversion of homocysteine to methi-onine by transfer of a methyl group from methyltet-rahydrofolate. This enzyme links the methylation pathway through synthesis of the methyl donor S-adenosyl methionine and the pathway in which purine and pyrim...
متن کاملFunctional characterization of a vitamin B12-dependent methylmalonyl pathway in Mycobacterium tuberculosis: implications for propionate metabolism during growth on fatty acids.
Mycobacterium tuberculosis is predicted to subsist on alternative carbon sources during persistence within the human host. Catabolism of odd- and branched-chain fatty acids, branched-chain amino acids, and cholesterol generates propionyl-coenzyme A (CoA) as a terminal, three-carbon (C(3)) product. Propionate constitutes a key precursor in lipid biosynthesis but is toxic if accumulated, potentia...
